Isolasi, Karakterisasi, Analisa Kimia Dan Deteksi BAPPT Bakteri Tanah Perakaran Padi Dari Rambut Siwi, Bali
Trhizosphere of rice plant is rich in organic compounds in the form of energy. Bacteria, especially “Plant growth promoting rhizobacteria (PGPR)” group liked rich habitat with energy resources to grow and compete. This study aims to determine the population, characterization and detection of bacteria as PGPR from rice rhizosphere that can be use as one of the tool to environmental rehabilitation of degraded land. The study used a selective media (Yema, Caceres, Mannitol Ashby, Pikovskaya) for isolation and population bacteria. Chemical analysis (IAA hormone, PMEase enzyme, ACC Deaminase) methods follow Mehboob, Tabatabai and Bremner, Penrose and Glick methods, whereas characterization (identifying) used molecular methods (analysis sequencing) with amplification of the 16S rRNA gene. Identification results found 10 species of bacteria namely Bacillus weihenstephanensis, lipoferum Azospirillum, Azotobacter crococcum, Stenotrophomonas maltophilia, Bacillus megaterium, Bacillus megaterium, Rhizobium sp., Rhizobium sp. were detected as PGPR. The highest population (41 CFU / g soil) of bacteria in rice rhizosphere is Azospirillum lipoferum RSB17. The bacteria also has the highest value on the production of IAA (5.596), saline resistant on 30g / L NaCl, and potential as a producer of ACC deaminase (grown a lot in the media DF + ACC). Bacillus megaterium RSB3 and RSB 9, Bacillus weihenstephanensis RSB18, and Azospirillum lipoferum RSB17 has the highest value of dissolved P and PMEase enzyme.
