Partial Cloning of Gene Encoding Mannanase from Indigenous Actinomycetes
Mannanase has been high potential for industry application, such as pulp bleaching in paper industry, production for functional food ingredient, increasing supply of feed, added aroma, taste and decreasing of consistency coffee extract, and application in the mining oil and gass. In this research, the gene encoding for a mannanase from five isolates of Actinomycetes belonged to Biotechnology Culture Collection (BTCC) was partial cloned by using pGEM-T Easy Vector System I (Promega) as vector cloning and Eschericia coli TOP10F (Invitrogen) was as a host for cloning. The DNA of each isolated bacteria were amplified by PCR using one set of degenerated primers. PCR result showed that the PCR product of about 250 bp which has high homology with gene sequences encoding mannanase gen from Streptomyces sp based on homology analysis with BLAST (Genbank). Further analysis will be conducted to get the whole of mannanase gene. Proceedings International Conference on Biotechnology.
